Formation and growth of adventitious roots from in vitro Angelica acutiloba Kitagawa tissue culture

Japanese touki is considered of high and prior value in many traditional remedies due to medicinal properties in the root system. In this study, root formation and growth of Japanese touki cultured in vitro were carried out in the dark condition of a culture room having temperature of 24 ± 2oC and RH of 65 ± 5%. Leaf blades from three different positions of the in vitro touki shoot were cultured either on basal mineral MS medium supplemented with Morel vitamins or Gamborg’s B5 medium. Percent of root formation was 100% as well as root fresh weight (132.7 mg explant-1) was the greatest when the second leaf blade was cultured on the Gamborg’s B5 medium. Percent of root formation, root number, root fresh weight and root fresh weight/explant fresh weight were the greatest when the leaf blade was cultured on the Gamborg’s B5 medium supplemented with 0.1 mg/l BA.

In the MS liquid medium including Morel vitamin, shaked at 100 ppm, and supplemented with different IBA concentrations (0.2, 0.5 or 1 mg/l), adventitious roots of in vitro touki plants grew and increased their biomass over time with the increase in IBA concentrations. On day 28, touki roots had the greatest fresh weight (2423.7 mg/vessel) in the medium supplemented with 1 mg/l IBA, almost 5 times higher than that (500 mg/vessel) at the first day of culture.
